Automated assay for the quantification of dissolved proteins in natural seawater samples

ثبت نشده
چکیده

An assay is described that permits the simultaneous determination of concentrations of total dissolved proteins and total "dissolved free amino acids" in natural seawater samples. It can be performed automatically even during field experiments using an AutoAnalyzer system or a modified amino acid analyzer. Except filtration, no further pretreatment of the samples is necessary. The determination is based on the reaction of o-phthaldialdehyde (OPA) with proteins or amino acids, respectively, after separating both components by a column chromatography step. For routine analysis of protein concentrations in natural seawater samples, the lower detection limit is at least 0.5 ngfsample. As the sample volume can be varied between 25 Jll and 2 000 J.Ll, concentrations below 0.5 J.Lg/1 can be analyzed. The determination of protein concentration in a single seawater sample takes about 30 minutes.

An automated photo-oxidation method for the determination of dissolved organic phosphorus in marine and fresh water TITRE

A segmented flow automated method with on-line photo-oxidation for the determination of dissolved organic phosphorus+soluble reactive phosphorus (DOP+SRP) in seawater and fresh water is described here. A low-power lamp was used for a compact, easy-to-handle and low-ozone-producing manifold. The influence of seawater matrix components was studied in detail using natural seawater and salt solutio...

Optimizing of microfunnel-supported liquid-phase microextraction for the extraction of chlorophenoxyacetic acids in seawater samples

Abstract In this study, microfunnel-supported liquid-phase microextraction method (MF-LPME) was optimized and applied for the determination of some chlorophenoxyacetic acids (CPAAs) from natural water samples. The extraction was performed with 540 μL toluene which was retained at the surface of the sample solution using an upside down microfunnel. After the extraction, toluene was moved in...

Real-time PCR assay for rapid detection and quantification of Vibrio aestuarianus in oyster and seawater: a useful tool for epidemiologic studies.

Because Vibrio aestuarianus is known to cause serious infections in Pacific oyster Crassostrea gigas, a real-time PCR assay was developed targeting the dnaJ gene of this bacterium. Only V. aestuarianus strains isolated from C. gigas mortality events in different geographic areas and the reference strain tested positive, whereas no amplification products was obtained with type strains belonging ...

Isolation and quantification of dissolved lignin from natural waters using solid-phase extraction and GC/MS.

Solid-phase extraction (SPE) was tested for the isolation of dissolved lignin from diverse natural waters (fresh, estuarine, and marine) in preparation for CuO oxidation. Capillary GC coupled to selected-ion monitoring mass spectrometry (SIM-MS) of CuO oxidation products provides the high sensitivity and precision required for the identification and quantification of trace levels of lignin in s...

Quantification Analysis of Dot Blot Assays for Human Immunodeficiency Virus Type 1 and 2 Antibodies

Objective Dot Blot (DB) assay provides highly specific results, but usually not reliable for quantification of antibody production. The need for a more objective DB assay to provide a better definition of the immune status, against HIV antigens, promoted this study to be done to develop a quantitative DB assay. Material and Methods Dot blot (DB) strips for antibodies directed to human immuno...
